Her paternal grandparents were Henry VI of Luxembourg and his wife, Beatrice of Avesnes. Her maternal grandparents were John I, Duke of Brabant and his second wife Margaret of Flanders.
Marie's siblings were John of Bohemia and Beatrice, Queen of Hungary. John was married to Elisabeth of Bohemia (1292–1330), among their children were, Charles IV, Holy Roman Emperor and Bonne of Bohemia. Beatrice married Charles I of Hungary but died while giving birth.
She was married in Paris to Charles IV of France following his divorce from his adulterous first wife, Blanche of Burgundy, on 21 September 1322. Blanche had given birth to two children, Philip and Joan, but both of them died young and so Charles needed a son and heir to carry on the House of Capet.
In 1323, Marie became pregnant but she later miscarried. Whilst pregnant again in March 1324, Marie was involved in a carriage accident, and seriously injured. Her child, a boy (Louis), was born prematurely, and died several hours later; Marie died on 26 March 1324, aged only nineteen, and was buried at Montargis in the Dominican church. Following her death Charles married Jeanne d'Évreux, who also failed to give birth to a son so the direct House of Capet was succeeded its branch, the House of Valois.
